Two modes of tissue regeneration in axolotl

Abstract

The axolotl is a premier model of tissue regeneration due to its ability to regenerate many different body parts including limb, tail and various internal organs including central nervous system. In this seminar, I will focus on what we have learnt from regeneration of two main appendages - limb and tail. While limb regeneration uses dedifferentiation of fibroblasts, we have found resident stem cell population (asomitic stem cell) in tail that is responsible for tail regeneration. Unlike limb where connective tissue and myogenic lineages strictly maintain their lineage boundaries, we have found that asomitic stem cells can contribute to both connective tissue and myogenic lineages during tail regeneration. Further investigation of asomitic stem cells suggests interesting differences between how limb and tail myogenesis proceeds during homeostatic development which provides further insights to their evolutionary and therapeutic importance. In the last ten minutes of my talk, I will discuss tissue clearing and mesoSPIM microscopy and their applications.
